ABSTRACT

The degrees of air sac lesions in turkeys with airsacculitis were standardized, associated with the presence of Enterobacteriaceae isolated from these organs. A total of 110 samples of air sacs from male turkeys with airsacculitis were collected and analyzed. During the slaughtering process, the sample collection was done using swabs and submitted to three storage methods (immediate, frozen, or pre incubated after freezing) for further comparison of their isolated efficiency. The bacterial genera of the family Enterobacteriaceae were identified biochemical series EPM, MILi and Simmons citrate. Bacterial growth occurred in 43.64% of samples. In this study, four patterns of aerossaculitis lesions were identified according to the pathological characteristics of air sacs. The frequencies of the Enterobacteriaceae isolated identified in the samples were: Escherichia coli, Citrobacter, Proteus, Edwardsiella, Morganell, Kluyvera, Salmonella and Klebsiella. Otherwise, it was observed that the levels already standardized as level three and four showed higher variety of genus. The immediate storage showed higher percentage of positivity at 41.82%, however, the pre incubated after freezing showed more efficiency in relation to the quantity of colonies.(AU)

ABSTRACT

The present study evaluated the dietary supplementation with essential oil of Lippia alba on the hemato-immunological parameters of Nile tilapia (Oreochromis niloticus) submitted to acute inflammation induced by carrageenin injection in the swim bladder. For a period of 45 days, 96 fish were divided in four treatments in triplicate, as follows: fish fed supplemented diet with essential oil of L. alba (4 mL kg-1 dry ration) injected with carrageenin; fish fed supplemented diet with cereal alcohol injected with carrageenin; fish fed unsupplemented diet with essential oil injected with carrageenin; fish fed unsupplemented diet and noninjected. Cortisol levels, erythrogram, leukogram and the inflammatory infiltrate were analyzed 6 h after inflammatory stimulus. Carrageenin-injected fish showed acute inflammatory reaction in the swim bladder characterized by higher infiltrate of neutrophils and monocytes. The circulating neutrophils number was significantly higher in fish fed L. alba when compared to other treatments. No difference in cortisol levels was found. For dose, time and administration form tested, supplementation with essential oil of L. alba did not present anti-inflammatory activity. On the other hand, influence of dietary supplementation was observed on the neutrophils number after induced aerocystitis highlighting its immunomodulatory characteristic.(AU)

ABSTRACT

Seasonal outbreaks of airsacculitis in China's poultry cause great economic losses annually. This study tried to unveil the potential role of Avian metapneumovirus (AMPV), Ornithobacterium rhinotracheale (ORT) and Chlamydia psittaci (CPS) in avian airsacculitis. A serological investigation of 673 breeder chickens and a case-controlled study of 430 birds were undertaken. Results showed that infection with AMPV, ORT, and CPS was highly associated with the disease. The correlation between AMPV and CPS were positively robust in both layers and broilers. Finally, we determined the co-infection with AMPV, ORT, and CPS was prevalent in the sampled poultry farms suffering from respiratory diseases and the outbreak of airsacculitis was closely related to simultaneous exposure to all three agents.

ABSTRACT

We describe here the mass death of predatory carp, Chanodichthys erythropterus, in Korea induced by plerocercoid larvae of Ligula intestinalis as a result of host manipulation. The carcasses of fish with ligulid larvae were first found in the river-edge areas of Chilgok-bo in Nakdong-gang (River), Korea at early February 2016. This ecological phenomena also occurred in the adjacent areas of 3 dams of Nakdong-gang, i.e., Gangjeong-bo, Dalseong-bo, and Hapcheon-Changnyeong-bo. Total 1,173 fish carcasses were collected from the 4 regions. To examine the cause of death, we captured 10 wondering carp in the river-edge areas of Hapcheon-Changnyeong-bo with a landing net. They were 24.0-28.5 cm in length and 147-257 g in weight, and had 2-11 plerocercoid larvae in the abdominal cavity. Their digestive organs were slender and empty, and reproductive organs were not observed at all. The plerocercoid larvae occupied almost all spaces of the abdominal cavity under the air bladders. The proportion of larvae per fish was 14.6-32.1% of body weight. The larvae were ivory-white, 21.5-63.0 cm long, and 6.0-13.8 g in weight. We suggest that the preference for the river-edge in infected fish during winter is a modified behavioral response by host manipulation of the tapeworm larvae. The life cycle of this tapeworm seems to be successfully continued as the infected fish can be easily eaten by avian definitive hosts.

ABSTRACT

In order to introduce an easily applicable, removable, painless and long-term drug delivery system for non-insulin dependent diabetes mellitus [NIDDM], hydroxyl propyl methyl cellulose with polyvinyl alcohol [HPMC-PVA] blend patches of metormin HC1 were evaluated in vitro and in vivo. A suitable patch of metformin 800mg with HPMC-PVA blend were used, following a three cycle freeze-thaw technique. Drug release kinetic profiles were performed in both patch and swim bladder. Albino mice were artificially generated as NIDDM mice by alloxan insertion i.p and after then treated with the therapeutic patch. Blood glucose was estimated by commercially available glucose kit based on glucose oxidase method. Drug release parameters from the patch and swim bladder were typical non-Fickian diffusion and both have the same kinetic constant, revealing its possible diffusion through stratum corneum. Hypoglycemia was observed in treatment of normal mice with TDDS of metformin HC1 within 4 hours i.e. 25 +/- 2.13mg/dl and within 16 hours in diabetic rats blood glucose level returned to normal level i.e. from 360 +/- 3.3mg/dl [NIDDM level] to 105 +/- 2.5 mg/dl [Normal level]. The TDS-patch has got the same kinetic simulation with that of swim-bladder, which might be a prediction for in vivo application. Here metformin was delivered to diabetic mice and has got significant anti-diabetic effect can be considered as a kind of patch for NIDDM just like wearing and taking off a hand watch because hypoglycaemia can be removed by just taking off the patch

ABSTRACT

The effects of swim bladder injection with thioglycolate, Escherichia coli lipopolysaccharide (LPS) and heat-inactivat ed Aeromonas hydrophila were assessed on hematological responses in pacu, Piaractus mesopotamicus (Characidae). A quantitative assessment was done on erythrocytes, thrombocytes e leucocytes at 6, 24, and 48 h pos-injection of the inflammatory agents and compared with fish injected with saline solution (control). Fish injected with inactivated A. hydrophila showed a reduction of erythrocytes and hemoglobin, whereas the hematocrit increased 6 h pos-injection. The results show that thioglycolate and LPS also induced a reduction on hemoglobin and an increase on the hematocrit. The thrombocytes count decreased 6 h post A. hydrophila injection, whereas increased 48 hours post LPS injection. The leukocytes count increased after 6 h post A. hydrophila injection, while the lymphocytes and PAS-positive granularleukocytes (PAS-LG) count decreased after 24 h post injection. In fish injected with thioglycolate or with LPS showed an increase in the LG-PAS counts when compared to A. hydrophila or control groups. The monocytes count was not affected by the different inflammatory agents.

ABSTRACT

Catfishes may have two sonic organs: pectoral spines for stridulation and swimbladder drumming muscles. The aim of this study was to characterize the sound production of the catfish Iheringichthys labrosus. The I. labrosus male and female emits two different types of sounds: stridulatory sounds (655.8 + 230 Hz) consisting of a train of pulses, and drumming sounds (220 + 46 Hz), which are composed of single-pulse harmonic signals. Stridulatory sounds are emitted during abduction of the pectoral spine. At the base of the spine there is a dorsal process that bears a series of ridges on its latero-ventral surface, and by pressing the ridges against the groove (with an unspecialized rough surface) during a fin sweep, the animal produce a series of short pulses. Drumming sound is produced by an extrinsic sonic muscle, originated on a flat tendon of the transverse process of the fourth vertebra and inserted on the rostral and ventral surface of the swimbladder. The sounds emitted by both mechanisms are emitted in distress situation. Distress was induced by manipulating fish in a laboratory tank while sounds were recorded. Our results indicate that the catfish initially emits a stridulatory sound, which is followed by a drumming sound. Simultaneous production of stridulatory and drumming sounds was also observed. The catfish drumming sounds were lower in dominant frequency than stridulatory sounds, and also exhibited a small degree of dominant frequency modulation. Another behaviour observed in this catfish was the pectoral spine locking. This reaction was always observed before the distress sound production. Like other authors outline, our results suggest that in the catfish I. labrosus stridulatory and drumming sounds may function primarily as a distress call.

ABSTRACT

Fish damage in hydroelectric stations has been extensively reported, but further investigations focusing on Brazilian migratory species, especially large fish, are needed. The present study describes swimbladder abnormalities in piapara Leporinus obtusidens, a migratory physoclistous species, collected downstream from the Funil Hydroelectric station dam, Minas Gerais, Brazil. The 117 specimens collected were dissected for swimbladder evaluation in the coelomic cavity. The abnormalities detected in 14 of the fish (12 percent) were swimbladder rupture with exteriorization (N = 12) and formation of sac-like projections (N = 2). The specimens that had swimbladder abnormalities weighed over 1300 g for both sexes. These abnormalities may be caused by bladder infections, however, inflammation process were not detected in these fish. The presence of anomalies in swimbladder suggests that the individuals were subjected to sudden depressurization, possibly related to passage through the turbines. Further comparative studies are needed to corroborate this hypothesis.

ABSTRACT

OBJECTIVETo explore the intervention of electro-acupuncture (EA) on cycloxygenase (COX) mRNA and protein expression induced by human recombination tumor necrosis factor-alpha (hrTNF).

METHODSAir sac model was established by implanted an autoclaved teflon chamber into back of rat (SPF grade). Ten days after modeling, 90 qualified rats, in them inflammation not detected, were randomly divided into three groups, the control group, the TNF model (model) group and the TNF +EA (TE) group, with 30 cases in each. To rats in the control group, 1 mL of saline was injected into the sac, but to those in the model and TE groups, 2 ng/mL hrTNF in a volume of 1 mL was used instead to induce local inflammatory responses. Immediately after then, EA (2 Hz, 5 mA and persistent waves) was applied to bilateral Quchi points (LI11) of rats in the TE group for 30 min. Fluid in the sac was drawn out at various time points (1, 5 and 24 h) after injection to measure the COX-2 mRNA expression by RT-PCR and COX-2 protein expression by Western blot.

RESULTSLevel of COX-2 mRNA expression in the control group was significantly different to that in the other two groups (P < 0.05 or P < 0.01). COX-2 mRNA expression at the 24th hour and COX-2 protein expression at the 1st hour were significantly lower in the TE group than those in the model group (P < 0.05 and P < 0.01) respectively.

CONCLUSIONEA could effectively intervene the mRNA and protein expression of COX-2 induced by hrTNF, which is possibly by way of pro-inflammatory cytokine regulating.

ABSTRACT

The Weberian apparatus is essentially a mechanical device improving audition, consisting of a double chain of ossicles joining the air bladder to the inner ear. Despite being one of the most notable complex systems of teleost fishes and the subject of several comparative, developmental and functional studies, there is still much controversy concerning the origin, evolution and homologies of the structures forming this apparatus. In this paper I provide a new insight on these topics, which takes into account the results of recent works on comparative anatomy, paleontology, and ontogeny as well as of a recent extensive phylogenetic analysis including not only numerous otophysan and non-otophysan extant otocephalans but also ostariophysan fossils such as Chanoides macropoma, Clupavus maroccanus, Santanichthys diasii, Lusitanichthys characiformis, Sorbininardus apuliensis and Tischlingerichthys viohli. According to the evidence now available, the Weberian apparatus of otophysans seems to be the outcome of a functional integration of features acquired in basal otocephalans and in basal ostariophysans, which were very likely not directly related with the functioning of this apparatus, and of features acquired in the nodes leading to the Otophysi and to the clade including the four extant otophysan orders, which could well have been the result of a selection directly related to the functioning of the apparatus.

ABSTRACT

The work was carried out on fifteen adult pigeons of both sexes. The gross morphological features of the air sacs were thoroughly investigated using Vinyl chloride acetate corrosion casts. It was found that the pigeon as an example of a powerful flying bird had 4 pairs of transparent air sacs [cervical, cranial thoracic, caudal thoracic and abdominal], in addition to an unpaired interclavicular sac. They were symmetrically occupying either side of the body cavity. The cervical sacs aerated the cervical vertebrae approximately from the 3rd or 4th as well as the first two thoracic vertebrae. The single interclavicular air sac was the most complicated among all air sacs in the pigeons. It occupied the ventral part of the thoracic inlet and overlay the cervical sacs laterally and connected with their cranial ends. Meanwhile, the interclavicular sac gave off extrathoracic humeral and axillary diverticula, which pneumatized the humeri and bones of the pectoral girdles. It was also observed that the cranial and caudal thoracic air sacs showed no bony communications, and never aerates bones. The paired abdominal sacs were larger than the other air sacs. They enclosed between their medial surfaces the intestinal tract, kidneys and the gonads. Also, each of them sent a diverticulum which ventilated the interior of the pelvic bone and the synsacrum. The obtained results of the air sacs in the pigeon were discussed with the findings given by previous authors in other species of birds

ABSTRACT

Light and electron microscopy coupled to Laser scanning confocal microscopy and immunohistology (for adhesion detection, revealed that in its development, the air pouch produced in mice, starts as a traumatic event in where is produced a subcutaneous air space surrounded by compressed pre-existing connective issue. It soon matures toward a well circunscribed air pouch, acquiring an epithelial-looking lining suported by newly synthesized connective tissue layers. We recognized three developmental stages and considered they should be taken in account in experimental designs making use of the air pouch model. The capability of the air pouch lining to express adhesion molecules, its easy manipulation and its situation as an isolated comportament turns the air pouch a good alternative model to the coelomatic cavities.

ABSTRACT

The metabolism of gas gland cells of the swimbladder epithelium is specialized for the production of acidic metabolites that are released into the blood stream and provoke an increase in gas partial pressure by reducing the effective gas-carrying capacity of the blood. In a subsequent step this initial increase in gas partial pressure is multiplied by back-diffusion of gas molecules from the venous to the arterial side in the countercurrent system, the rete mirabile. Thus, gas partial pressures of up to several hundred atmospheres can be generated in the swimbladder. Measurements of metabolic end products and analysis of the formation of 14C02 from [1-14(superscription) C] glucose and [6-14(superscription) C] glucose revealed that the acidic metabolises are lactic acid, produced in the glycolytic pathway, and also C02, formed in the pentose phosphate shunt. C02 easily enters the blood stream by diffusion. The release of protons from isolated gas gland cells, however, is highly dependent on the extracellular sodium concentration. This sodium dependence can in part be blocked by addition of amiloride, indicating that a Na+/ H+ exchanger is involved in the release of protons. A significant decrease in the rate of proton secretion in the presence of the carbonic anhydrase inhibitor ethoxzolamide indicates that the second major route for the release of protons includes carbonic anhydrase activity and the diffusion of C02.
